MMP-11 Colorimetric Cell-Based ELISA Kit

Assay Genie offers a cell-based ELISA kit for the quantitative measurement of Matrix Metalloproteinase-11 (MMP-11) levels, using a colorimetric detection method read at 450 nm. The kit is a convenient, lysate-free, high throughput and sensitive assay designed to detect the MMP-11 protein expression profile directly in cultured cells, and can be used to measure relative amounts of MMP-11 as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on MMP-11.

Reactivity has been validated for human, mouse and rat samples. The assay uses an indirect ELISA format in which MMP-11 is captured by MMP-11-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the conjugated HRP enzyme then catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density for further normalisation. The dynamic range supports measurements above 5000 cells, and the kit is intended to be stored at 4 degrees C.

MMP-11 (also known as Stromelysin-3, gene symbol MMP11) plays a key role in tissue remodelling and is implicated in diseases such as cancer and arthritis, belonging to the peptidase M10A family. By accurately measuring MMP-11 levels, researchers can gain insight into disease progression, potential therapeutic targets and treatment efficacy monitoring.
