Description

From Assay Genie, this Cell-Based ELISA is designed for the qualitative determination of MKP1 (Mitogen-activated protein kinase phosphatase-1) protein expression in cultured human cells. Detection is colorimetric, measured at 450 nm, with a dynamic range of > 5000 Cells. The kit is supplied in a 96-well microplate format and is intended for use with adherent cells, with a stated storage stability of 4°C for 6 Months.

The assay follows an indirect ELISA format in which MKP1 is captured by MKP1-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, enabling normalization of absorbance values to cell amounts. The kit can be used for measuring relative amounts of MKP1 in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MKP1.

MKP-1 (DUSP1) is a non-receptor, dual-specificity phosphoprotein phosphatase that specifically inactivates mitogen-activated protein (MAP) kinase by dephosphorylation. Its dysregulation has been linked to cancer, inflammatory disorders, and neurodegenerative conditions, making it a valuable target for therapeutic and biomarker research.
