MKP-1/2 (Phospho-Ser296/318)Colorimetric Cell-Based ELISA Kit

An In-Cell ELISA from Assay Genie designed for the detection of phosphorylated MKP-1/2 (Phospho-Ser296/318) protein levels in cultured cells. The kit is reactive with Human, Mouse and Rat samples and uses a Colorimetric 450 nm detection method. It is a convenient, lysate-free, high-throughput and sensitive assay that can detect MKP-1/2 protein phosphorylation and expression profile in cells, and can be applied to measure the influence of various treatments, inhibitors (such as siRNA or chemicals) or activators on MKP-1/2 phosphorylation.

The assay uses an indirect ELISA format, in which MKP-1/2 (Phospho-Ser296/318) is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing the target absorbance values. Following colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, with results normalized to cell amounts to adjust for plating differences. The kit is suitable for use with adherent cells.
