MKP-1/2 Colorimetric Cell-Based ELISA Kit

Assay Genie provides a cell-based ELISA kit for the qualitative detection of MKP-1 and MKP-2 protein expression in cultured cells. The colorimetric assay uses a 96-well microplate format with detection at 450 nm and a dynamic range of > 5000 cells, and it is reactive with human, mouse, and rat samples. It is intended for use with adherent cells and is suited to high-throughput, large-scale studies.

The kit employs a lysate-free, indirect ELISA format in which MKP-1/2-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. The assay allows researchers to measure relative MKP-1/2 levels in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators.

MKP-1 and MKP-2 (gene symbols DUSP1/4) are dual-specificity phosphoprotein phosphatases that play a key role in regulating cellular processes such as proliferation and differentiation, and their dysregulation has been linked to cancer, inflammation, and neurodegenerative disorders. The kit should be stored at 4°C and is stable for 6 months under those conditions.
