MKK3 Colorimetric Cell-Based ELISA Kit

This Cell-Based ELISA Kit from Assay Genie is designed for the sensitive and specific detection of mitogen-activated protein kinase kinase 3 (MKK3) levels in cell lysates. It provides a convenient, lysate-free, high-throughput assay for measuring the relative amounts of MKK3 in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MKK3.

Qualitative determination of MKK3 concentration is achieved by an indirect ELISA format, in which MKK3 is captured by MKK3-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density.

Reactivity is reported for Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells and detection at 450 nm. The target is MKK3, encoded by the gene MAP2K3 (Gene ID: 5606, UniProt ID: P46734), a dual specificity protein kinase that acts as a key regulator of stress response pathways and plays a role in cell proliferation, differentiation, and apoptosis. The kit is intended for use with Adherent Cells, is configured in a 96-Well Microplate format, and is stored at 4 °C with a stated stability of 6 months.
