The blue variant accumulates in mitochondria as a function of mitochondrial mass and is excited by the violet 405 nm laser, with emission collected through a 460/50 nm bandpass filter on standard flow cytometers. The dried reagent is reconstituted in DMSO, diluted into pre-warmed culture medium, and applied to cells at 37 degrees C with 5% CO2 for 15-30 minutes before washing and fixation with 1-4% formaldehyde. Store the lyophilized dye refrigerated and protected from light; once reconstituted, the working solution should be used promptly or aliquoted and frozen to limit freeze-thaw loss of signal intensity. Common applications include comparing mitochondrial content across drug-treated hematopoietic populations, sorting live mitochondria-rich subsets, and pairing with viability dyes such as 7-AAD or DAPI for dead-cell exclusion in multi-color panels. The probe also supports time-course studies of mitophagy, oxidative stress response, and metabolic reprogramming in cancer and stem cell models.
