Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the sensitive detection of MET phosphorylated at tyrosine 1356 in cell lysates, supporting research into the receptor tyrosine kinase MET, also known as hepatocyte growth factor receptor (HGFR), and its role in cellular processes such as proliferation, survival, and motility. Phosphorylation of MET at tyrosine 1356 is associated with activation of downstream signaling pathways involved in tumor development and progression.

The assay uses an indirect ELISA format in which Met (Phospho-Tyr1356) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies catalyse a colorimetric reaction read at 450 nm. The kit is described as lysate-free, high throughput, and suitable for adherent cells, with a stated dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density for further normalization. Reactivity is reported for Human, Mouse, and Rat samples.

MET (UniProt ID: P08581, Gene ID: 4233) is a receptor tyrosine kinase of approximately 85,745 Da that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor/HGF ligand, activating downstream cascades including RAS-ERK, PI3 kinase-AKT, and PLCgamma-PKC. The encoded preproprotein is proteolytically processed to generate alpha and beta subunits linked via disulfide bonds to form the mature receptor. Mutations, amplification, and overexpression of the MET gene are associated with papillary renal cell carcinoma, hepatocellular carcinoma, and various head and neck cancers.
