Description

A fluorometric cell-based ELISA kit from Assay Genie designed to monitor Met (Phospho-Tyr1234) protein phosphorylation and the expression profile of the target in cells. The assay is lysate-free and high throughput, and uses an indirect ELISA format in which target-specific primary antibodies capture Met (Phospho-Tyr1234), followed by Dye 1-conjugated and Dye 2-conjugated secondary antibodies that enable fluorometric detection. It is suited to measuring the relative amounts of phosphorylated Met in cultured cells and to screening the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on Met phosphorylation.

Reactivity is reported for Human, Mouse and Rat samples. Two normalization approaches are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and an antibody against the non-phosphorylated counterpart of Met is included so that the RFU values obtained for non-phosphorylated Met can be used to normalize the RFU values for phosphorylated Met. The reported dynamic range is > 5000 Cells, and plates are read at Ex/Em 651/667 (Dye 1) and 495/521 (Dye 2). Storage stability is stated as 4°C for 6 Months. The target is Phospho-Met (Tyr1234), with the MET gene (Gene ID: 4233, UniProt ID: P08581) encoding the hepatocyte growth factor receptor, a receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor/HGF ligand and regulates processes including proliferation, scattering, morphogenesis and survival.
