Description

Designed for the accurate detection of phosphorylated Merlin (Phospho-Ser518) levels in cell lysates, this colorimetric cell-based ELISA kit from Assay Genie offers high sensitivity and specificity for research into cell signaling pathways. Merlin, also known as NF2 (Neurofibromin 2), is a tumor suppressor protein that plays a critical role in cell growth regulation and cytoskeletal organization. Phosphorylation of Merlin at Serine 518 has been linked to various cancers, including meningiomas and schwannomas, making it a valuable biomarker for cancer research and drug development.

The assay is a convenient, lysate-free, high-throughput method that can detect Merlin protein phosphorylation and expression profiles in cultured cells, making it suitable for measuring relative amounts of phosphorylated Merlin as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Merlin phosphorylation. Detection is achieved via an indirect ELISA format in which Merlin (Phospho-Ser518)-specific primary antibodies are captured by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density.

The kit is reactive with human, mouse, and rat samples and is intended for use with adherent cells, with a stated dynamic range of > 5000 cells. Database references listed include Gene ID 4771, UniProt ID P35240, and Gene Symbol NF2.
