Description

This cell-based ELISA kit from Assay Genie is designed to detect MEK2 (MAP2K2) protein expression directly in cultured cells, using a colorimetric readout at 450 nm. The assay is lysate-free and works with adherent cells, with reactivity established for human, mouse, and rat samples. It offers a dynamic range of > 5000 cells and is supplied in a microplate format for high-throughput workflows.

Detection is carried out by an indirect ELISA approach: MEK2 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Because the readout is qualitative, two normalisation strategies are built in. A monoclonal antibody specific for human GAPDH serves as an internal positive control to normalise target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences.

The kit can be applied to measure the relative amounts of MEK2 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MEK2 expression. MEK2, also known as Mitogen-Activated Protein Kinase Kinase 2, is described by the manufacturer as a dual-specificity protein kinase of the STE7 family, phosphorylated and activated by Raf and Mos kinases, that phosphorylates a Thr and a Tyr residue in the activation loop of ERK2 and ERK3, acting as a component of MAP kinase signal transduction pathways involved in mitogen growth factor signalling. The recommended storage condition is 4 °C with a stated stability of 6 months.
