Description

This cell-based ELISA kit from Assay Genie is designed for the detection and quantification of phosphorylated MEF2A at serine 408 in cell lysates or tissue homogenates. The kit uses a colorimetric detection method measured at 450 nm and is reactive with human, mouse, and rat samples, with a dynamic range supporting analysis of more than 5,000 cells.

It is a lysate-free, high-throughput assay that measures relative MEF2A phosphorylation and expression profiles in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MEF2A phosphorylation. Detection is achieved through an indirect ELISA approach, in which phosphorylation-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition.

Two normalization methods are provided: an included monoclonal antibody specific for human GAPDH serves as an internal positive control for absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. Phosphorylation of MEF2A at serine 408 is known to play a critical role in regulating gene expression and cellular processes involved in cardiac development, muscle differentiation, and neuronal plasticity, and its dysregulation has been implicated in cardiovascular disorders, neurodegenerative diseases, and cancer.
