Description

A colorimetric, cell-based ELISA kit from Assay Genie is designed for the measurement of MED1 protein levels in cell lysates and tissue samples. The assay uses a colorimetric detection method at 450 nm and shows reactivity with human and mouse samples. It is supplied in a 96-well microplate format and is intended for adherent cell samples.

Working through an indirect ELISA format, the kit captures MED1 using MED1-specific primary antibodies and detects the bound antibody with HRP-conjugated secondary antibodies, which catalyze a colorimetric reaction upon substrate addition. The stated dynamic range is greater than 5000 cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density and adjust for plating differences. The kit is stored at 4°C with a stated stability of 6 months.

MED1 is a subunit of the CRSP complex required for efficient activation by SP1, and it also functions as a coactivator for nuclear receptors such as the thyroid hormone receptor. The protein is associated with p53-dependent apoptosis and is described as essential for adipogenesis, with two splice-variant isoforms reported.
