Description

A cell-based colorimetric ELISA kit from Assay Genie designed to detect phosphorylation of MCM4 at serine 54 in adherent cells. The assay targets MCM4 (Phospho-Ser54) and is reactive with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. Detection is performed colorimetrically at 450 nm using HRP-conjugated secondary antibodies in an indirect ELISA format, and the kit is described as offering high sensitivity and specificity.

Provided as a convenient, lysate-free, high-throughput workflow, the kit enables measurement of relative MCM4 phosphorylation in cultured cells and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MCM4 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric readout, Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences.

MCM4 is a mini-chromosome maintenance protein essential for the initiation of eukaryotic genome replication and forms part of the pre-replication complex. The MCM hexameric complex possesses DNA helicase activity, and phosphorylation by CDC2 has been reported to reduce that activity as well as chromatin binding of the complex. The kit is classified under the Cell Cycle research area and as a Phospho-specific sub type.
