MAST4 Colorimetric Cell-Based ELISA

Assay Genie offers a Colorimetric Cell-Based ELISA for the qualitative detection of MAST4 protein expression in cultured cells, with read-out at 450 nm on a 96-well microplate. The kit is lysate-free and designed for adherent cell samples, with reactivity against human and mouse targets.

Detection uses an indirect ELISA workflow: MAST4-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to drive a colorimetric reaction on substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, while Crystal Violet whole-cell staining provides a measure of cell density so that absorbance values can be corrected to plating differences. The assay supports a dynamic range of > 5000 Cells and is shipped for storage at 4°C with a 6-month stability.

MAST4 is a member of the microtubule-associated serine/threonine protein kinase family, classified within the AGC Ser/Thr protein kinase superfamily, with 5 human isoforms produced by alternative splicing. It has been linked to cellular signalling processes including cell proliferation, migration and survival, and its dysregulation has been associated with cancer, neurological disorders and autoimmune diseases. The target corresponds to Gene ID 375449 and UniProt entry O15021, with the gene located at chromosomal position 5q12.3.
