MART-1 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed for the detection of MART-1 protein levels in cells, cell lysates and tissue samples. The assay uses an indirect ELISA format in which MART-1 is captured by MART-1-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. The kit is described as a convenient, lysate-free, high-throughput and sensitive assay that can be used to measure the relative amounts of MART-1 in cultured cells and to screen for the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on MART-1 expression.

Detection is colorimetric, read at 450 nm, and the kit is reactive with human samples. MART-1, also known as Melan-A, is a melanocyte-specific protein expressed in melanocytes and melanoma cells, so monitoring its levels can support studies of melanoma progression, metastasis and response to treatment. Because the assay is qualitative, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used after the HRP colorimetric read-out to determine cell density and adjust for plating differences. Storage is at 4°C with a stability of 6 months.
