Description

This cell-based ELISA kit from Assay Genie provides a convenient, lysate-free, high-throughput method for detecting MARK3 protein expression profiles in cultured cells. The assay uses a colorimetric detection method at 450 nm in a 96-well microplate format and is reactive with human, mouse, and rat samples. It is suited for measuring relative MARK3 levels in cultured adherent cells and for screening the effects of various treatments, inhibitors, or activators on MARK3 expression.

Qualitative determination of MARK3 concentration is achieved through an indirect ELISA format: MARK3-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following the colorimetric measurement, a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalised to cell amounts. The dynamic range exceeds 5000 cells.

MARK3 (MAP/microtubule affinity-regulating kinase 3) is a ubiquitous serine/threonine kinase of the CAMKL family, recorded under UniProt ID P27448 and Gene ID 4140. The kit is stored at 4 °C with a stated stability of six months.
