Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of MAPKAPK5 levels in cell lysates and tissue homogenates. It uses a colorimetric detection method at 450 nm, is reactive with human and mouse samples, and has a stated dynamic range of > 5000 cells. The kit is provided in a 96-well microplate format and is suitable for adherent cells.

The assay uses an indirect ELISA format in which MAPKAPK5 is captured by specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to enable colorimetric readout upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining method is provided to determine cell density so that absorbance values can be normalized to cell amounts. The kit can be used to measure relative MAPKAPK5 expression in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on MAPKAPK5.

MAPKAPK5, also known as PRAK, is a serine/threonine protein kinase involved in stress response and cell proliferation. It is activated through phosphorylation by MAP kinases including ERK, p38-alpha, and p38-beta in response to cellular stress and proinflammatory cytokines, and participates in signaling pathways that regulate cell growth, differentiation, and apoptosis. The kit is stored at 4 °C with a stated stability of 6 months.
