Description

Assay Genie provides a cell-based ELISA kit designed for the specific detection of phosphorylated MAPKAPK2 at Thr334 in cultured cells. The assay uses a colorimetric readout at 450 nm in a 96-well microplate format and is validated for human, mouse, and rat samples. It offers a dynamic range of > 5000 cells, making it appropriate for measuring relative levels of phosphorylated MAPKAPK2 in adherent cell cultures.

The kit operates on an indirect ELISA principle, in which target-specific primary antibodies capture phosphorylated MAPKAPK2 at Thr334, and HRP-conjugated secondary antibodies then catalyse the colorimetric reaction upon substrate addition. Because of the qualitative nature of the cell-based ELISA, two normalisation approaches are included: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step allows absorbance values to be normalised to cell density. Researchers can use the kit to assess MAPKAPK2 phosphorylation status in response to various treatments, inhibitors, or activators, supporting studies of inflammation, stress response, and cell signalling in cancer and related disease models.

MAPKAPK2 is a Ser/Thr protein kinase belonging to the MAPKAPK family and is phosphorylated and activated by p38 MAP kinase in response to cytokines, stress, and chemotactic factors, with HSP27 identified as one of its in vivo substrates. The phospho-specific kit is grouped within the Epigenetics and Nuclear Signaling research area and targets the Thr334 phospho-site.
