Description

Assay Genie's cell-based ELISA kit is designed for the qualitative detection of MAP3K7 phosphorylation at serine 439 in cultured cells. The assay uses a colorimetric method read at 450 nm and an indirect ELISA format in which target-specific primary antibodies are detected with HRP-conjugated secondary antibodies, producing a measurable color reaction upon substrate addition. The kit is provided in a 96-well microplate format and is described as lysate-free, high throughput, and sensitive.

Reactivity is reported for human, mouse, and rat samples, with a stated dynamic range of > 5000 cells. The kit can be used to measure relative levels of phosphorylated MAP3K7 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MAP3K7 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining method is recommended to determine cell density after the colorimetric measurement.

MAP3K7, also known as TAK1, is a serine/threonine kinase of the MLK family that mediates signal transduction induced by TGF beta and morphogenetic protein (BMP). It controls a variety of cell functions including transcription regulation and apoptosis, and also activates MAPK8/JNK and MAP2K4/MKK4, playing a role in the cellular response to environmental stresses.
