Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative measurement of MAP2K7 protein phosphorylation at Ser271 in cultured cells. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures the target, and an HRP-conjugated secondary antibody catalyses a colorimetric reaction at 450 nm upon substrate addition. A monoclonal antibody against human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step may be used to determine cell density after the colorimetric readout.

The reagents are reactive with human, mouse and rat samples and are suitable for adherent cells, with a dynamic range above 5000 cells. The workflow is lysate-free and high-throughput, allowing screening of the effects that various treatments, siRNA or chemical inhibitors, and activators have on MAP2K7 phosphorylation.

MAP2K7, also known as MEK7, is a dual-specificity protein kinase of the STE7 family that activates JNK1 and JNK2 within the MAPK signalling cascade. Phosphorylation at Ser271 plays a crucial role in its activation and downstream biological functions, and dysregulation of MAP2K7 signalling has been implicated in cancer, inflammatory disorders and neurodegenerative conditions, making its phosphorylation status a valuable readout in cell death and signalling research.
