Description

This colorimetric, cell-based ELISA from Assay Genie provides a high-throughput, lysate-free workflow for measuring MAP2K7 protein levels directly in cultured cells. The assay uses an indirect ELISA format: target-specific primary antibodies capture MAP2K7 within fixed cells, and HRP-conjugated secondary antibodies bind the Fc region of those primaries, catalysing a colorimetric reaction that is read at 450 nm. The kit is reactive with human, mouse and rat samples, is intended for adherent cell cultures, and has a dynamic range of more than 5000 cells per well. It is supplied in a microplate format and is stored at 4 °C with a stated stability of 6 months.

Because the read-out is qualitative, two normalisation strategies are built in. A monoclonal anti-GAPDH antibody is included to act as an internal positive control for plate-to-plate comparison, and a Crystal Violet whole-cell stain can be used to determine cell density so that absorbance values can be adjusted for plating differences. The kit is suitable for profiling relative MAP2K7 expression as well as screening the effects of treatments, inhibitors (including siRNA or chemical compounds) and activators on MAP2K7 levels in cells.

MAP2K7, also referred to as MEK7, is a dual-specificity protein kinase of the STE7 family that activates JNK1 and JNK2 by phosphorylating a threonine and a tyrosine residue in their activation loops. It sits within the MAPK signalling cascade, where it contributes to the regulation of cell growth, proliferation and differentiation, and its dysregulation has been linked to cancer, inflammatory disorders and neurodegenerative disease. The target is identified in NCBI as Gene ID 5609 and in UniProt as O14733.
