MAFF Transcription Factor Activity Assay

The MAFF Transcription Factor Activity Assay from Assay Genie measures the functional DNA-binding activity of MAFF (v-maf musculoaponeurotic fibrosarcoma oncogene homolog F), a small Maf transcription factor that heterodimerizes with cap'n'collar (CNC) family partners such as NRF2 and binds Maf recognition element (MARE) sequences to drive antioxidant and detoxification gene expression.

The plate is pre-coated with a double-stranded MARE consensus oligonucleotide; functionally active MAFF in prepared nuclear and cell lysates binds the immobilized sequence and is quantified by immunodetection with a MAFF-specific antibody. Researchers studying oxidative stress responses, Nrf2-MAFF pathway activation, glutathione S-transferase induction, or heme oxygenase-1 expression can use this ELISA as a quantitative alternative to EMSA or Western blot analysis.

A minimal volume of nuclear extract is loaded per well, and reagents should be stored refrigerated with aliquoted components to limit freeze-thaw cycles.
