Description

Designed for the accurate detection of MAF protein levels in cell cultures, this Cell-Based ELISA Kit from Assay Genie offers a convenient, lysate-free, high-throughput and sensitive assay for measuring the relative amounts of Maf in cultured cells. It can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on Maf expression. Qualitative determination of Maf concentration is achieved through an indirect ELISA format in which Maf-specific primary antibodies capture the target, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP enzyme to catalyse a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalisation methods are supplied: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining is used to determine cell density after the colorimetric measurement.

The assay is intended for use with adherent cells and is reactive with Human, Mouse and Rat samples. Detection is colorimetric, read at 450 nm, with a dynamic range of > 5000 Cells. The plate format is a 96-well microplate and the kit should be stored at 4 °C, where it remains stable for 6 months. MAF is a DNA-binding leucine zipper-containing transcription factor of the bZIP family (Maf subfamily) that can act as a transcriptional activator or repressor depending on context, and is involved in processes including embryonic lens fiber cell development, chondrocyte terminal differentiation during endochondral bone development, modulation of T-cell apoptosis through interaction with MYB and decreased BCL2 expression, and activation of IL4 expression in T helper 2 (Th2) cells.
