Description

From Assay Genie, this in-cell ELISA is designed for the sensitive detection of MAEA in adherent cell cultures. Operating as a lysate-free, high-throughput assay in a 96-well microplate, it provides qualitative measurement of MAEA expression through an indirect ELISA format in which MAEA-specific primary antibodies are paired with HRP-conjugated secondary antibodies.

The kit is reactive with human, mouse and rat samples and has a stated dynamic range of > 5000 cells. It is intended for research in the cell cycle area, and can be used to measure relative MAEA levels in cultured cells as well as to screen the effects of treatments, inhibitors or activators on MAEA expression. Detection is colorimetric, with absorbance read at 450 nm, and the kit is stored at 4°C with a stated stability of 6 months.

Normalization of the colorimetric signal is supported by two approaches: a monoclonal anti-human GAPDH antibody supplied as an internal positive control, and Crystal Violet whole-cell staining to determine cell density after the readout. The MAEA target, also known as Macrophage erythroblast attacher, is an integral membrane protein encoded by the MAEA gene (Gene ID: 10296; UniProt ID: Q7L5Y9). It mediates attachment of erythroid cells to mature macrophages, contributes to erythroblastic island formation and the development of mature macrophages, and associates with F-actin to regulate actin distribution.
