Description

Designed as a Cell-Based ELISA from Assay Genie, this kit targets LKB1 phosphorylated at Thr189 and is intended for the detection of phosphorylated LKB1 protein levels in cell lysates. It is reactive with human and mouse samples and uses a colorimetric detection method measured at 450 nm. The kit is described as a lysate-free, high throughput and sensitive assay that detects LKB1 protein phosphorylation and expression profile in cells, with a dynamic range of > 5000 Cells.

Qualitative determination of LKB1 (Phospho-Thr189) concentration is achieved by an indirect ELISA format. The target is captured by LKB1 (Phospho-Thr189)-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted. The kit can be used for measuring the relative amounts of phosphorylated LKB1 in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on LKB1 phosphorylation.

LKB1, also known as Liver Kinase B1, is a key regulator of cell metabolism, growth, and polarity. Phosphorylation of LKB1 at Thr189 is essential for its activation and plays a role in signalling pathways related to energy balance and cell proliferation. Dysregulation of LKB1 signalling has been implicated in diseases such as cancer, diabetes, and neurodegenerative disorders. Database information provided with the kit lists Gene Symbol STK11, Gene ID 6794, UniProt ID Q15831, and OMIM entries 175200, 273300, and 602216.
