Description

A colorimetric cell-based ELISA kit from Assay Genie designed for the detection and quantification of phosphorylated LKB1 at serine 428 in cell lysates. The assay uses an indirect ELISA format in which LKB1 (Phospho-Ser428) is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies catalyse a colorimetric reaction read at 450 nm. It is reactive with human and mouse samples and operates across a dynamic range of > 5000 cells.

The kit is lysate-free and intended for adherent cells, allowing measurement of the relative amounts of phosphorylated LKB1 in cultured cells and screening of the effects that various treatments, inhibitors such as siRNA or chemicals, or activators have on LKB1 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density, so that absorbance values can be normalised to cell amounts and plating differences adjusted.

LKB1 (Liver kinase B1) is a serine/threonine kinase of the CAMKL family encoded by STK11 (Gene ID: 6794, UniProt ID: Q15831). It acts as a tumour suppressor that helps control cell structure, polarity, apoptosis and energy homeostasis, and exists in a cytosolic complex with the pseudokinase STRAD and the MO25 scaffold protein to activate AMPK and related kinases. Phosphorylation at Ser428 modulates its activity, making this kit relevant to studies of cellular signalling in cancer, metabolic disorders and other diseases where LKB1 signalling is implicated.
