Description

A colorimetric cell-based ELISA kit from Assay Genie designed for the in-cell detection of LKB1 protein, the serine/threonine kinase encoded by the STK11 gene. The assay uses an indirect ELISA format in which LKB1 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction read at 450 nm. It is described as a lysate-free, high-throughput and sensitive assay for measuring relative amounts of LKB1 in cultured adherent cells, and is suited to screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators.

The kit reports reactivity with human and mouse samples and a dynamic range of > 5000 cells. Two normalisation methods are provided: an anti-GAPDH monoclonal antibody serves as an internal positive control for the target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density after the HRP/substrate readout, allowing absorbance values to be normalised to cell amounts. Storage is specified at 4 °C with a stability of 6 months.

LKB1 functions as a tumour suppressor that helps control cell structure, polarity, apoptosis and energy homeostasis, acting through a cytosolic complex with the pseudokinase STRAD and the MO25 scaffold protein to activate AMPK and related kinases. Dysregulation of LKB1 has been implicated in cancer, diabetes and neurodegenerative disorders, and the kit is positioned for research into LKB1 biology and potential therapeutic targets.
