LIMK1 Colorimetric Cell-Based ELISA

This In-Cell ELISA Kit from Assay Genie provides a colorimetric method for detecting LIM kinase 1 (LIMK1) levels in cell lysates. LIMK1 is a key regulator of actin cytoskeleton dynamics and is involved in cellular processes including cell migration, invasion, and metastasis. The kit is reactive with Human, Mouse, and Rat samples and is intended for use with adherent cell samples.

The assay is supplied in a 96-Well Microplate format with colorimetric detection at 450 nm and has a dynamic range of > 5000 Cells. It is a convenient, lysate-free, high-throughput and sensitive assay for measuring LIMK1 protein expression profiles in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on LIMK1.

Qualitative determination of LIMK1 concentration is achieved by an indirect ELISA format, in which LIMK1 is captured by LIMK1-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be normalised to cell amounts.
