Description

Assay Genie provides a Cell-Based ELISA Kit for the qualitative determination of Lamin A/C (Phospho-Ser22) in cultured adherent cells. Detection is colorimetric, measured at 450 nm, and the workflow is lysate-free and high-throughput, allowing researchers to measure relative phosphorylation levels in cells as well as screen the influence of treatments, inhibitors (such as siRNA or chemicals), or activators on Lamin A/C phosphorylation.

The assay uses an indirect ELISA format in which Lamin A/C (Phospho-Ser22) is captured by phospho-specific primary antibodies and revealed by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step may be used to determine cell density, so that absorbance values can be normalized to cell amounts and plating differences adjusted.

The kit reacts with Human, Mouse and Rat samples and has a dynamic range of > 5000 Cells. It supports research into nuclear assembly, chromatin organization and gene expression regulation, where dysregulation of Lamin A/C phosphorylation has been implicated in cancer, muscular dystrophy and premature aging syndromes. The target is the LMNA gene product (Gene ID: 4000; UniProt ID: P02545), classified as a Phospho sub-type.
