Lamin A/C Colorimetric Cell-Based ELISA Kit

Designed for the quantitative detection of Lamin A/C levels in cell lysates and tissue samples, this cell-based ELISA kit from Assay Genie provides a convenient, lysate-free, high-throughput, and sensitive assay for characterising Lamin A/C protein expression in cultured cells. The assay is suitable for measuring the relative amounts of Lamin A/C in cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Lamin A/C. Reactivity is reported for Human, Mouse, and Rat, and the kit is intended for use with adherent cells.

Detection uses a colorimetric method measured at 450 nm, with a dynamic range of > 5000 cells. Qualitative determination of Lamin A/C concentration is achieved via an indirect ELISA format: Lamin A/C is captured by Lamin A/C-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalisation methods are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density and adjust for plating differences.
