Description

The Assay Genie Ku80/XRCC5 (Phospho-Thr714) Colorimetric Cell-Based ELISA Kit is a cell-based assay designed to detect Ku80/XRCC5 protein phosphorylation and expression in cells. The kit uses an indirect ELISA format, in which Ku80/XRCC5 (Phospho-Thr714) is captured by target-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalyzing a colorimetric reaction at 450 nm upon substrate addition.

Reactive with human samples, the kit has a dynamic range of "> 5000 Cells" and is suitable for adherent cells. It can be used to measure the relative amounts of phosphorylated Ku80/XRCC5 in cultured cells, as well as to screen for the effects that various treatments, inhibitors, or activators have on Ku80/XRCC5 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and the Crystal Violet whole-cell staining method may be used to determine cell density.

Ku80, the 80-kilodalton subunit of the Ku complex (also known as ATP-dependent DNA helicase II), functions in the repair of DNA double-strand breaks by non-homologous end joining and in the completion of V(D)J recombination events. Phosphorylation at Thr714 plays a role in regulating its function in DNA damage response processes, and dysregulation of KU80 phosphorylation has been implicated in cancer and genetic disorders, highlighting its significance as a potential therapeutic target and biomarker.
