KRT37/38 Colorimetric Cell-Based ELISA Kit

Assay Genie offers a colorimetric cell-based ELISA kit designed for the qualitative measurement of KRT37 and KRT38 protein expression in cultured cells. The assay uses an indirect ELISA format in which the targets are captured by KRT37/38-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. It is described as lysate-free, high-throughput, and sensitive, and can be used to measure relative KRT37/38 levels in cells as well as to screen for the effects of treatments, inhibitors (such as siRNA or chemicals), or activators.

Detection is performed colorimetrically at 450 nm in a 96-well microplate format, with a dynamic range of "> 5000 Cells." The kit is intended for use with Human samples and adherent cells and is stored at 4°C for 6 Months. KRT37 and KRT38 are keratin proteins that contribute to the structural integrity of epithelial cells, and their dysregulation has been linked to various skin disorders, making them relevant biomarkers for keratin biology and skin-related research. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density after colorimetric measurement.

The kit includes KRT37/38 primary antibodies, HRP-conjugated anti-rabbit and anti-mouse IgG secondary antibodies, 10X TBS, blocking buffer, wash buffer, quenching buffer, substrate, stop solution, crystal violet solution, and adhesive plate seals. A microplate reader able to measure absorbance at 450 nm (and optionally at 595 nm for Crystal Violet cell staining) is required but not provided.
