Description

From Assay Genie, this Cell-Based ELISA kit is designed for the sensitive detection of intracellular KPBB protein in cell-based assays. The kit targets KPBB, also known as PHKB (phosphorylase b kinase regulatory subunit beta, UniProt ID: Q93100), and is reactive with human and mouse samples, supporting research in metabolism and cellular signaling pathways. It is supplied in a 96-well microplate format and is intended primarily for use with adherent cells.

Detection is colorimetric, measured at 450 nm, and uses an indirect ELISA format in which KPBB is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies. The protocol is lysate-free and high-throughput, with a dynamic range of > 5000 Cells, and allows qualitative measurement of relative KPBB levels in cultured cells as well as assessment of the effects of treatments, inhibitors, or activators. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step is provided for cell density determination. Storage is at 4°C with 6 months stability.

Beyond applications in drug discovery, toxicology, and basic research, the kit is relevant to studies of glycogen metabolism: defects in PHKB are associated with glycogen storage disease type 9B. The target protein has a reported molecular weight of 124 kDa and is encoded at chromosomal location 16q12.1.
