KNTC2 Colorimetric Cell-Based ELISA

The Assay Genie Cell-Based ELISA Kit is designed for analyzing expression levels of Kinetochore protein KNTC2 in cell lysates. KNTC2 is a key player in chromosome segregation during cell division, making it a vital target for studying cell cycle regulation and identifying potential therapeutic targets for cancer and other diseases. The assay is a lysate-free, high-throughput and sensitive format that detects the KNTC2 protein expression profile in cells. It can be used for measuring relative amounts of KNTC2 in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on KNTC2. Qualitative determination of KNTC2 concentration is achieved by an indirect ELISA format, where KNTC2 is captured by KNTC2-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

The kit uses a colorimetric detection method read at 450 nm and is reactive with human and mouse samples. It has a dynamic range of > 5000 Cells and is configured for adherent cells in a microplate format. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density. Storage is at 4°C for 6 months.
