KLF10/11 Transcription Factor Activity Assay

KLF10 and KLF11 are Sp/Krüppel-like transcription factors that bind GC-rich promoter elements and regulate cell proliferation, differentiation, and TGF-β signaling. This Assay Genie DNA-binding ELISA uses an immobilized double-stranded oligonucleotide probe carrying the KLF10/11 consensus binding site to capture active factor from nuclear and cell lysates, with bound factor detected by a KLF10/11-specific antibody. The assay produces a quantitative readout of DNA-binding activity without gel-shift electrophoresis, and the kit is validated for reactivity with human, mouse, and rat samples. Each microplate kit contains reagents for 96 individual reactions, and nuclear extracts should be freshly prepared before the assay; aliquot samples and freeze at -80 °C, avoiding repeated freeze-thaw cycles to preserve binding activity. The kit is intended for research use only in studies of transcriptional regulation in oncology, fibrosis, and metabolic disease research.
