KLF Transcription Factor Activity Assay

The KLF (Krüppel-like factor) family comprises zinc-finger transcription regulators that bind GC-rich and CACCC-box DNA elements to direct gene programs in erythropoiesis, epithelial differentiation, adipogenesis, and immune cell maturation. This DNA-binding immunoassay from Assay Genie quantifies active KLF protein engaging its consensus sequence within nuclear and whole-cell lysates, using biotinylated double-stranded oligonucleotides immobilized on streptavidin-coated microplate wells and an antibody-based colorimetric readout.

Sample volume: 10 uL of nuclear extract per well. Standard assay workflow finishes in approximately 3.5 hours at room temperature after extract preparation. Reagents are stored at -20 °C and should be aliquoted into single-use portions to limit freeze-thaw cycles; diluted working buffers may be kept refrigerated at 4 °C and protected from light for short-term use.
