Kir6.2 (Phospho-Thr224)Colorimetric Cell-Based ELISA Kit

A Colorimetric Cell-Based ELISA Kit from Assay Genie enables detection and quantification of phosphorylated KIR6.2 protein in cell lysates and tissue samples. The kit provides a lysate-free, high-throughput, and sensitive assay suitable for measuring relative amounts of phosphorylated Kir6.2 in cultured cells, as well as for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Kir6.2 phosphorylation.

The assay uses an indirect ELISA format in which Kir6.2 (Phospho-Thr224) is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody. The HRP enzyme catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, and absorbance values can be normalised to cell amounts to adjust for plating differences.

The kit is reactive with Human and Mouse samples, with a dynamic range of > 5000 cells and detection by colorimetric measurement at 450 nm. It is intended for use with adherent cells. KIR6.2 is a key component of ATP-sensitive potassium channels that regulates cellular excitability and insulin secretion, with phosphorylation at Thr224 modulating channel activity.
