Description

A cell-based colorimetric ELISA kit from Assay Genie designed for the quantitative detection of KIF20A in cell lysates and culture supernatants, using adherent cells as the sample type. The assay is configured for human reactivity and read at a Colorimetric 450 nm detection wavelength, with a stated dynamic range of > 5000 Cells and a storage stability of 4°C/6 Months.

The kit uses an indirect ELISA format in which KIF20A is captured by KIF20A-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction on substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method can be used after the colorimetric measurement to determine cell density, allowing absorbance values to be normalised to cell amounts. The procedure is lysate-free and supports measuring relative KIF20A amounts in cultured cells as well as screening the effects of treatments, inhibitors, or activators on KIF20A.

Database identifiers associated with the target are Gene ID: 10112, UniProt ID: O95235, OMIM: 605664, and Unigene: Hs.718626. KIF20A (kinesin family member 20A) is described in the source as a mitotic kinesin required for chromosome passenger complex (CPC)-mediated cytokinesis, with roles in PLK1 recruitment to the central spindle and retrograde RAB6-regulated transport of Golgi membranes and associated vesicles along microtubules, and its dysregulation has been implicated in cancer progression.
