Description

This colorimetric, cell-based ELISA kit from Assay Genie is specifically designed for the accurate and sensitive detection of phosphorylated KIF11 (EG5) at Thr926 in cell lysates. The assay targets KIF11/Eg5 (Phospho-Thr926) and is reactive with human samples, with a stated dynamic range of > 5000 Cells. Detection is performed by colorimetric measurement at 450 nm, and the kit supports research in cell cycle biology, including studies of cell division, cancer biology, and potential therapeutic interventions, as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on KIF11/Eg5 phosphorylation.

The kit uses an indirect ELISA format in which KIF11/Eg5 (Phospho-Thr926) is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. Because the cell-based ELISA is qualitative, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density for normalization of absorbance values.

KIF11 (EG5) is a motor protein of the kinesin-like family (UniProt P52732; NCBI Gene ID 3832; molecular weight 119,159 Da) that is essential for cell division and plays a critical role in spindle formation and chromosome segregation during mitosis. Phosphorylation of KIF11 at Thr926 is reported to regulate its function and has been implicated in cancer progression and drug resistance.
