Description

Assay Genie's colorimetric cell-based ELISA kit enables qualitative detection of Keratin 8 phosphorylated at Serine 73 in cultured cells, using an indirect ELISA format. The assay is reactive with human and mouse samples and employs a colorimetric detection method read at 450 nm, with a dynamic range of > 5000 cells.

Designed as a lysate-free, high-throughput and sensitive assay, the kit detects Keratin 8 protein phosphorylation and expression profiles in cells, allowing measurement of the relative amounts of phosphorylated Keratin 8 in cultured cells and screening of the effects that various treatments, inhibitors, or activators have on Keratin 8 phosphorylation. Keratin 8 (Phospho-Ser73)-specific primary antibodies capture the target, while HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining method may be used to determine cell density.

Keratin 8 (KRT8, Gene ID: 3856) is a type II cytoskeletal keratin involved in maintaining the structural integrity of epithelial cells. Phosphorylation of keratins at specific sites affects their organization, assembly dynamics, and interaction with signaling molecules, and dysregulation of keratin 8 has been implicated in liver diseases, inflammatory disorders, and cancers.
