Keratin 8 Colorimetric Cell-Based ELISA Kit

A cell-based ELISA kit from Assay Genie enables qualitative measurement of Keratin 8 protein expression in cultured adherent cells without requiring a lysis step. The kit uses an indirect ELISA format in which Keratin 8 is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences.

The assay is suited to measuring the relative amounts of Keratin 8 in cultured cells and to screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on Keratin 8 expression. The dynamic range exceeds 5000 cells and the kit is reactive with human and mouse samples. The plate format is a 96-well microplate, with reagent storage specified at 4°C for 6 months. Keratin 8 is a type II cytoskeletal keratin that contributes to the structural integrity of epithelial cells, and abnormal levels have been linked to liver diseases, inflammatory disorders, and certain cancers.
