Description

From Assay Genie, this cell-based ELISA kit is designed for the precise quantification of keratin 20 levels in cell lysates and tissue homogenates. The colorimetric detection at 450 nm provides accurate and reproducible results, suitable for researchers studying keratin biology and related diseases. The kit is reactive with human, mouse, and rat samples, has a dynamic range of more than 5000 cells, and is intended for use with adherent cells.

Working as a convenient, lysate-free, high throughput and sensitive assay, it can detect the keratin 20 protein expression profile in cells. It is suitable for measuring the relative amounts of keratin 20 in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on keratin 20. Qualitative determination is achieved by an indirect ELISA format: keratin 20 is captured by keratin 20-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, with results normalised to cell amounts to adjust for plating differences. Storage is recommended at 4 °C with 6 months stability.
