Description

Designed for the detection of Keratin 18 phosphorylated at serine 33, this Cell-Based ELISA Kit from Assay Genie provides a lysate-free, high-throughput indirect ELISA format for assessing Keratin 18 protein phosphorylation and expression profiles in cultured cells. Phospho-specific and anti-target primary antibodies capture the analyte, while HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm upon substrate addition, enabling qualitative measurement of Keratin 18 (Phospho-Ser33) concentration.

The assay is reactive with Human, Mouse, and Rat samples, uses adherent cells, and is positioned within the Cell Cycle research area. It supports screening for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Keratin 18 phosphorylation. Two normalization strategies are included: an anti-GAPDH monoclonal antibody serves as an internal positive control for target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance readings to be adjusted for plating differences.

The kit has a stated dynamic range of greater than 5000 cells. The underlying target, KRT18 (Gene ID: 3875; UniProt: P05783), encodes a type I cytoskeletal keratin expressed in single-layer epithelial tissues, and phosphorylation of Keratin 18 at serine 33 has been linked to cellular processes including cell migration, proliferation, and apoptosis, making this kit a relevant tool for research in cancer, liver disease, and related disorders.
