Description

From Assay Genie, this Cell-Based ELISA Kit enables quantitative measurement of Keratin 18 protein levels in cultured cells using a colorimetric detection method. The kit is a lysate-free, high-throughput, and sensitive assay designed to determine relative Keratin 18 expression in adherent cells and to screen the effects of various treatments, inhibitors such as siRNA or chemicals, and activators.

Detection uses an indirect ELISA format in which Keratin 18 is captured by target-specific primary antibodies and bound by HRP-conjugated secondary antibodies. Signal is developed through an HRP-catalysed colorimetric reaction and read at 450 nm. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance values, while a Crystal Violet whole-cell stain can be used to determine cell density for further normalisation. The assay has a dynamic range of > 5000 Cells and has been validated for reactivity with Human, Mouse, and Rat samples.

Keratin 18 is a type I cytoskeletal intermediate filament protein that contributes to the structural integrity of epithelial cells and is expressed in single-layer epithelial tissues of the body. The kit is supplied in a 96-well microplate format, is intended for Cell Cycle research applications, and is stored at 4°C with a stated stability of 6 Months.
