Description

Assay Genie offers this cell-based ELISA kit for the qualitative detection of Keratin 15 in adherent cell cultures. The kit is a lysate-free, high-throughput assay designed to measure relative Keratin 15 protein expression in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Keratin 15. The assay uses an indirect ELISA format in which Keratin 15 is captured by Keratin 15-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

The kit is reactive with Human, Mouse, and Rat samples and employs a colorimetric detection method measured at 450 nm. It is supplied in a 96-well microplate format with a dynamic range of > 5000 Cells. For storage, the kit is stable at 4°C for 6 Months. An Anti-GAPDH monoclonal antibody is included to serve as an internal positive control for normalising target absorbance values, and following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method can be used to determine cell density, allowing results to be normalised to cell amounts and adjusting for plating differences.

Keratin 15 (gene symbol KRT15) is a type I cytoskeletal keratin that contributes to the structural integrity of epithelial cells, particularly in the skin and hair follicles. The protein has a molecular weight of 49kDa, is located on chromosome 17q21.2, and is associated with cellular components in the cytosol and nucleus, with molecular functions in protein binding and as a structural constituent of the cytoskeleton. It is involved in biological processes including epidermis development and keratinization. The UniProt ID is P19012.
