Description

From Assay Genie, this Cell-Based ELISA kit enables the precise measurement of Keratin 14 levels in cell lysates, tissue homogenates, and other biological samples. The assay uses a colorimetric detection method read at 450 nm and is reactive with Human, Mouse, and Rat samples, with adherent cells as the recommended sample type.

The kit is a convenient, lysate-free, high-throughput and sensitive format that qualitatively determines Keratin 14 protein expression in cultured cells, including the screening of treatments, inhibitors such as siRNA or chemicals, and activators. Detection is by an indirect ELISA approach: Keratin 14 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to plating differences.

Supplied in a microplate format with a dynamic range of greater than 5000 cells, the kit is intended to be stored at 4 °C with a stated stability of 6 months. Keratin 14, encoded by the KRT14 gene (UniProt P02533), is a type I cytoskeletal keratin of approximately 52 kDa that plays a critical role in maintaining the structural integrity of epithelial cells in tissues such as skin and hair follicles.
