Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of Keratin 10 protein expression in cultured cells and for the accurate, reliable detection of Keratin 10 levels in cell culture supernatants. The assay uses an indirect ELISA format in which Keratin 10 is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition. The procedure is lysate-free and supports high-throughput screening of treatments, inhibitors (such as siRNA or chemicals), or activators on Keratin 10.

Read using a colorimetric method at 450 nm, the assay includes a monoclonal anti-human GAPDH antibody as an internal positive control for normalising target absorbance values, and provides a Crystal Violet whole-cell staining step to determine cell density so that absorbance values can be normalised to cell amounts. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. It is supplied in a 96-Well Microplate format, is intended for use with adherent cells, and is stored at 4°C with a stability of 6 months.

Keratin 10 is a type I cytoskeletal keratin encoded by the KRT10 gene (Gene ID: 3858; UniProt ID: P13645), with a molecular weight of 58,827 Da. It plays a key role in the formation and maintenance of the skin barrier, making it an important biomarker for studying skin disorders, wound healing, and epithelial differentiation. The kit is suited to research in dermatology and skin biology, where accurate measurement of Keratin 10 can support investigations into skin conditions and potential therapeutic approaches.
