Description

From Assay Genie, this Cell-Based ELISA Kit is a lysate-free, high throughput and sensitive assay designed to detect the KAP1 protein expression profile in cultured cells. Using an indirect ELISA format, KAP1 is captured by KAP1-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction upon substrate addition, with detection measured at 450 nm. The kit is intended for adherent cell samples.

Reactivity covers human, mouse, and rat samples, with a stated dynamic range of > 5000 cells. The kit can be used to measure relative KAP1 levels in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. Storage is at 4°C for 6 months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density so absorbance values can be adjusted for plating differences.

KAP1 is the protein product of the PRKAR1B gene (UniProt ID: P31321), a cAMP-dependent protein kinase type I-beta regulatory subunit with a molecular weight of 43,073 Da. It is a regulatory subunit of the cAMP-dependent protein kinases involved in cAMP signalling in cells.
