JunD Colorimetric Cell-Based ELISA Kit

Designed for in-cell measurement of JunD, this Cell-Based ELISA Kit from Assay Genie enables detection of JunD protein expression in cultured cells without the need for lysate preparation. The kit targets JunD, a transcription factor of the AP-1 family that plays a role in regulating gene expression, cell proliferation and apoptosis, and it is reactive with human, mouse and rat samples. It is intended for use with adherent cells, cell lysates and tissue homogenates. Detection is colorimetric, read at 450 nm, with a stated dynamic range of > 5000 Cells. The assay uses an indirect ELISA format, with target-specific primary antibodies recognised by HRP-conjugated secondary antibodies that catalyse a colorimetric reaction upon substrate addition.

Supplied in a 96-Well Microplate format, the kit supports high-throughput workflows and is suited to measuring the relative amounts of JunD in cultured cells, as well as screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be normalised to cell number and plating differences can be adjusted. The kit is stored at 4 °C with a stability of 6 months and is aimed at research in Epigenetics and Nuclear Signaling.

JunD is encoded by the JUND gene (Gene ID: 3727; UniProt ID: P17535), a member of the JUN family and a functional component of the AP-1 transcription factor complex. The protein binds DNA as a dimer, and dysregulation of JunD has been linked to cancer, inflammatory disorders and autoimmune conditions. The protein has a reported molecular weight of 35,174 Da, with the human orthologue located at chromosomal position 19p13.2.
