Description

This Cell-Based ELISA kit from Assay Genie is designed for the qualitative measurement of Inositol-Tetrakisphosphate 1-Kinase 1 (ITPK1) protein expression in cultured cells. It uses a 96-Well Microplate format with a colorimetric detection method read at 450 nm, shows reactivity with human and mouse samples, and has a dynamic range of greater than 5000 cells per well. The kit is intended for use with adherent cells.

The assay is based on an indirect ELISA format in which ITPK1 is captured by ITPK1-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme then catalyzes a colorimetric reaction upon substrate addition. Because the Cell-Based ELISA is qualitative in nature, two normalization tools are included: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step allows absorbance values to be adjusted to cell density. The kit is lysate-free and suited to high-throughput workflows, and can be used to measure relative ITPK1 levels in cultured cells or to screen the effects of treatments, inhibitors (such as siRNA or chemicals), and activators on ITPK1.

ITPK1 is a kinase involved in inositol phosphate metabolism, with roles in regulating cell growth, differentiation, and signaling pathways. The target gene is catalogued under NCBI Gene ID 3705 and UniProt entry Q13572 (OMIM 601838), and the encoded protein, Inositol-tetrakisphosphate 1-kinase, has a listed molecular weight of 35,632 Da. The kit is stored at 4 °C with a stated stability of 6 months.
